ABO2202 is an investigational mRNA-based therapeutic designed to encode a bispecific antibody targeting Epithelial Cell Adhesion Molecule (EpCAM) and CD3. Developed by Ruijin Hospital at Shanghai Jiao Tong University School of Medicine, the drug is formulated as a nanoparticle for intraperitoneal administration. Its mechanism involves the in vivo production of bispecific antibodies that bridge EpCAM-expressing tumor cells (common in gastric cancer and peritoneal metastases) with CD3-positive T cells, thereby inducing T-cell mediated cytotoxicity against the cancer cells. It is currently being evaluated in clinical trials for advanced gastric cancer with peritoneal metastasis and malignant ascites, often in combination with systemic chemotherapy.
