ABP-1119 is a novel small molecule multi-kinase inhibitor developed by AB Pharma for the treatment of pancreatic cancer. It targets several tyrosine kinases involved in tumor progression, specifically EGFR, HER2, ALK, and BTK. In preclinical studies, ABP-1119 demonstrated potent enzymatic inhibition with IC50 values in the low nanomolar range (0.9 nM for EGFR and ALK, 2.1 nM for BTK, and 4.8 nM for HER2) and significant tumor growth inhibition in pancreatic cancer xenograft models (Mia-Paca-2), outperforming erlotinib. The compound has shown a favorable safety profile in Ames, hERG, and maximum tolerated dose (MTD) studies, along with good metabolic stability in human microsomes (T1/2 = 2.5hr).
