Drug intelligence / Profile preview
ABREV001 is a small molecule pan-mutant KRAS inhibitor developed by Agastiya Biotech LLC that targets multiple KRAS mutations including G12C, G12D, G12V, and G13D. It is described as a first-in-class inhibitor that binds to the GTP pocket in mutant KRAS, preventing downstream signaling and activating the proteasomal degradation pathway to trigger apoptotic cell death. The drug is intended for the treatment of KRAS-mutated solid tumors, including non-small cell lung cancer (NSCLC), colorectal cancer (CRC), and pancreatic cancer. Preclinical studies reported a 95% tumor volume reduction in vivo with minimal toxicity. While the company's lead program AB001 has advanced to Phase 2, ABREV001 remains a distinct preclinical asset focused on the GTP-bound (active) conformation of KRAS.
