Overview

Abrilumab is a fully human monoclonal IgG2 antibody that targets the α4β7 integrin heterodimer. By binding with high affinity to α4β7 integrin, it blocks its interaction with mucosal addressin cell adhesion molecule-1 (MAdCAM-1), which is selectively expressed in gut endothelium and upregulated in patients with inflammatory bowel disease (IBD). This blockade inhibits the migration of leukocytes into gastrointestinal tissue, thereby reducing inflammation associated with conditions such as ulcerative colitis and Crohn's disease. Abrilumab was developed for the treatment of inflammatory bowel diseases but has not been approved for any indication[1][3][5][6].
