ABS-106 is a preclinical-stage bispecific T-cell engager (BiTE) being developed by Amberstone Biosciences for the treatment of solid tumors. It is built upon the proprietary T-MATE (Tumor Microenvironment Activated Therapeutic) technology platform, which is designed to create conditionally active immunotherapeutics. The drug is engineered to remain relatively inactive in systemic circulation and healthy tissues, becoming fully potent only within the acidic or protease-rich tumor microenvironment. This selectivity aims to mitigate the systemic toxicities often associated with T-cell engagers, such as cytokine release syndrome (CRS). Mechanistically, ABS-106 simultaneously binds to CD3 on T cells and a tumor-associated antigen (TAA) on cancer cells, facilitating the recruitment and activation of cytotoxic T cells to induce tumor cell lysis. While the specific TAA target has not been publicly disclosed, the molecule is part of a broader pipeline of conditionally active bispecifics targeting solid malignancies.
