Overview

ABSK043 is an oral, small molecule inhibitor of programmed cell death ligand 1 (PD-L1), developed for the treatment of various advanced solid tumors, including non-small cell lung cancer (NSCLC), gastric/gastroesophageal junction cancer, metastatic unresectable urothelial carcinoma, melanoma, breast cancer, and endometrial cancer. By blocking PD-L1 from interacting with its receptor PD-1 on immune cells, ABSK043 aims to enhance the body's immune response against tumor cells. Unlike monoclonal antibody-based checkpoint inhibitors targeting the same pathway, ABSK043 offers the convenience of oral administration and potentially lower immunogenic risk. The drug is currently in Phase II clinical trials both as monotherapy and in combination with other agents such as furmonertinib for EGFR-mutated NSCLC[2][3][5][7].
