ABSK044

ABSK044 is an orally administered, small molecule inhibitor of the colony-stimulating factor 1 receptor (CSF1R), currently under development by Abbisko Therapeutics. By selectively binding to and inhibiting CSF1R, ABSK044 disrupts the signaling pathway essential for the survival and proliferation of macrophages, including tumor-associated macrophages (TAMs), and osteoclasts. This mechanism is particularly relevant in tenosynovial giant cell tumors (TGCT), which are characterized by the overexpression of CSF1, leading to the recruitment of CSF1R-expressing cells that drive tumor growth. Beyond TGCT, ABSK044 is being investigated for its potential to modulate the immunosuppressive tumor microenvironment in various advanced solid tumors, potentially enhancing the efficacy of other immunotherapies.
