ABSK111 is a potent, selective, and CNS-penetrable small molecule inhibitor of the epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 2 (HER2). Developed by Abbisko Therapeutics, it is designed to target a broad spectrum of mutations, including EGFR exon 20 insertions, atypical mutations, and extracellular mutations, as well as HER2 mutations. Notably, ABSK111 demonstrates high selectivity for mutant forms over wild-type EGFR, potentially reducing the severe adverse events associated with wild-type inhibition. Its ability to cross the blood-brain barrier makes it a candidate for treating central nervous system malignancies such as glioblastoma multiforme (GBM), in addition to non-small cell lung cancer (NSCLC).
