Overview

ABSK112 is a novel, next-generation small molecule inhibitor developed by Abbisko Therapeutics for the treatment of cancers driven by EGFR exon 20 mutations and HER2 alterations. It demonstrates improved selectivity over wild-type EGFR and strong brain penetration. Preclinical studies have shown potent inhibition of proliferation in multiple EGFR exon 20 mutation cell lines and superior efficacy compared to other inhibitors such as mobocertinib. Additionally, ABSK112 acts as a selective and CNS-penetrant HER2 inhibitor, supporting its potential use in treating HER2-positive cancers with brain metastases. The drug is currently being evaluated in Phase 1 clinical trials for metastatic non-small cell lung cancer (NSCLC), with oral administration as capsules[1][2][3][5][6].
