Overview

ABT‑288 is a highly selective and potent small molecule antagonist of the histamine H3 receptor. It was developed as an investigational therapy for cognitive impairment associated with disorders such as Alzheimer's disease and schizophrenia. By antagonizing the H3 receptor in the central nervous system, ABT‑288 enhances neurotransmitter release (notably acetylcholine and dopamine), which is thought to improve cognitive function. Preclinical studies demonstrated procognitive effects in animal models relevant to both Alzheimer's disease and schizophrenia. However, clinical trials in patients with mild-to-moderate Alzheimer's dementia and cognitive impairment associated with schizophrenia did not show efficacy over placebo; development was discontinued after these results. The drug showed generally good safety and tolerability profiles during clinical testing[1][5][6].
