Overview

ABT-301 is an oral small molecule inhibitor of histone deacetylases 1, 2, and 3 (HDAC1/2/3) being developed by Anbogen Therapeutics. It is designed to modulate the tumor microenvironment and enhance immune responses in patients with proficient mismatch repair (pMMR) or non-microsatellite instability-high (non-MSI-H) metastatic colorectal cancer (mCRC). Preclinical studies indicate that ABT-301 can convert immunologically "cold" tumors into "hot" tumors by promoting CD8+ cytotoxic T cell infiltration, enhancing antigen presentation, and inhibiting myeloid-derived suppressor cells. In clinical evaluations, ABT-301 has shown a favorable safety profile without the neutropenia or cardiac toxicity often associated with other HDAC inhibitors. It is currently being studied in a Phase 1/2 trial as part of a triplet combination therapy with tislelizumab and bevacizumab.
