ABT-700-PBD is an antibody-drug conjugate (ADC) composed of the MET-targeted monoclonal antibody ABT-700 (also known as h224G11) conjugated to a pyrrolobenzodiazepine (PBD) dimer payload. The antibody component, ABT-700, is a highly selective antagonist of the MET receptor tyrosine kinase (hepatocyte growth factor receptor), which is frequently overexpressed or activated in various malignancies, including colorectal cancer (CRC). The PBD payload is a potent DNA-crosslinking agent that induces irreversible DNA damage and cell death upon internalization into target cells. ABT-700-PBD was developed to address adaptive resistance in CRC, specifically targeting MET-positive cells that emerge following the loss of LGR5 expression during treatment with LGR5-targeted therapies. Preclinical data indicate that ABT-700-PBD exhibits dose-dependent cytotoxicity and significant tumor regression in patient-derived xenograft models, particularly when used in combination with other cancer stem cell-targeted agents.
