ABT-747 is a typographical error that occasionally appears in scientific literature in place of ABT-737, a first-in-class, small-molecule BH3-mimetic developed by Abbott Laboratories. ABT-737 is a potent inhibitor of the anti-apoptotic B-cell lymphoma 2 (Bcl-2) family proteins, specifically targeting Bcl-2, Bcl-xL, and Bcl-w with subnanomolar affinity, while sparing Mcl-1 and Bfl-1. By mimicking BH3-only proteins, it disrupts the interaction between anti-apoptotic and pro-apoptotic proteins (such as Bax and Bak), thereby triggering the intrinsic mitochondrial apoptotic pathway. Although ABT-737 demonstrated robust preclinical efficacy in various hematological malignancies (such as acute myeloid leukemia and lymphoma) and small-cell lung cancer, its clinical development was limited by poor aqueous solubility and lack of oral bioavailability. This led to the development of second-generation orally active analogs like navitoclax (ABT-263) and venetoclax (ABT-199).
