Overview

ABT-767 is an orally available small molecule inhibitor of the nuclear enzymes poly(ADP-ribose) polymerase 1 and 2 (PARP-1 and PARP-2). By selectively binding to these enzymes, ABT-767 inhibits their activity, thereby preventing the repair of damaged DNA via the base excision repair pathway. This mechanism leads to increased DNA damage in cancer cells, particularly those with deficiencies in homologous recombination repair such as BRCA1 or BRCA2 mutations. The drug was primarily developed for use in advanced solid tumors including high-grade serous ovarian cancer, fallopian tube carcinoma, and primary peritoneal carcinoma. Clinical studies have shown that patients with BRCA mutations or homologous recombination deficiency are more likely to respond to ABT-767[1][2][4][8].
