ABT-869 (linifanib) + paclitaxel is an investigational combination therapy evaluated primarily in oncology, especially breast carcinoma and non-small cell lung cancer (NSCLC). ABT-869 (linifanib) is a selective, orally bioavailable **receptor tyrosine kinase (RTK) inhibitor** targeting vascular endothelial growth factor (VEGF) and platelet-derived growth factor (PDGF) receptor families, as well as mutant constitutively active FLT3 and KIT kinases. Its primary mechanism is antiangiogenic and antiproliferative action by inhibiting tumor cell signaling and vasculature. Paclitaxel is an established **microtubule inhibitor** that stabilizes microtubules, preventing cell division. Preclinical and phase 1/2 studies demonstrate that the combination enhances antitumor efficacy, particularly in breast and lung cancers, compared to either agent alone, with reported partial responses in metastatic breast cancer and tolerability concerns primarily related to the ABT-869 dose[1][2][3][5][6][7][8][9].
