Overview

ABTL0812 + gemcitabine + nab-paclitaxel is a combination regimen under clinical investigation for cancer, specifically pancreatic cancer. ABTL0812 is a first-in-class, orally administered small molecule developed by AbilityPharma that induces cytotoxic autophagy in cancer cells by a dual mechanism: (1) inhibition of the Akt/mTORC1 pathway via upregulation of TRIB3 pseudokinase (itself driven by activating PPARα and PPARγ), and (2) induction of endoplasmic reticulum (ER) stress, leading to activation of the unfolded protein response. These converge to drive robust autophagy and cancer cell death. Gemcitabine is a nucleoside analog (antimetabolite) and standard chemotherapy, while nab-paclitaxel is a nanoparticle albumin-bound formulation of the antitubulin agent paclitaxel, providing enhanced delivery and antineoplastic activity. This combination is primarily being evaluated in advanced or metastatic pancreatic cancer in clinical trials, with rationale based on synergistic anticancer effects and the ability of ABTL0812 to overcome chemoresistance and augment standard chemotherapy efficacy[1][3][6].
