ABX-1431 (also known as Lu AG06466) is a first-in-class, potent, and selective small-molecule inhibitor of monoacylglycerol lipase (MGLL). Developed by Abide Therapeutics and later acquired by Lundbeck, it is designed to modulate the endocannabinoid system by preventing the breakdown of 2-arachidonoylglycerol (2-AG), the most abundant endocannabinoid in the brain. By increasing 2-AG levels, ABX-1431 enhances signaling at cannabinoid receptors, which helps regulate neurotransmitter release and modulate neural circuits that are overactive in certain neurological conditions. The drug was primarily evaluated in Phase 2 clinical trials for Tourette Syndrome and Chronic Motor Tic Disorder, though development for these indications was discontinued. It has also been investigated for other central nervous system disorders, including pain and spasticity associated with multiple sclerosis.
