ABX-196 is a synthetic glycolipid agonist of invariant Natural Killer T cells (iNKT), formulated in a liposomal preparation and administered intramuscularly. It is designed to activate iNKT cells, which are key regulators of immune responses with both immunomodulatory and antineoplastic activities. Upon administration, ABX-196 binds to iNKT cells via the CD1d receptor, leading to their activation and subsequent secretion of cytokines that stimulate various immune cell types—including NK cells, T cells, macrophages, and B cells—thereby enhancing anti-tumor immunity. Preclinical studies have shown that ABX-196 can reduce tumor growth and increase survival in models of hepatocellular carcinoma (HCC), both as monotherapy and in combination with checkpoint inhibitors such as nivolumab. Clinical trials have demonstrated safety, tolerability, potent iNKT activation in healthy volunteers (Phase 1), and promising clinical benefit signals in HCC patients when combined with nivolumab[2][3][5][8].
