ABX900 is a bispecific Centyrin biologic developed by Aro Biotherapeutics, currently in late-stage lead optimization for the treatment of advanced non-small cell lung cancer (NSCLC). Centyrins are a proprietary class of small, structurally simple, and highly stable proteins derived from the human tenascin-C fibronectin type III domain. ABX900 is engineered to simultaneously target and inhibit both the epidermal growth factor receptor (EGFR) and the mesenchymal-epithelial transition factor (MET). This dual-targeting approach is specifically designed to overcome resistance to existing EGFR inhibitors, such as osimertinib (Tagrisso), which often occurs through MET amplification or bypass signaling. The molecule's small size and high solubility potentially offer advantages in tissue penetration and formulation compared to traditional bispecific antibodies.
