ABY-025 is a small engineered Affibody molecule (approximately 7–7.6 kDa) that specifically targets the human epidermal growth factor receptor type 2 (HER2). It is used as a diagnostic radiopharmaceutical for noninvasive imaging of HER2 expression in tumors using PET or SPECT modalities when labeled with isotopes such as gallium Ga 68 or indium In 111. Upon intravenous administration, radiolabeled ABY-025 binds with high affinity to HER2-expressing tumor cells, enabling detection and quantification of HER2 status in cancers such as metastatic breast cancer and gastroesophageal junction cancer. This facilitates patient stratification for HER2-targeted therapies and assessment of therapeutic response. The molecule was developed by protein engineering to improve its biochemical properties and minimize immunogenicity[1][5][8].
