ABY-071 is a radioligand therapy (RLT) candidate developed by Affibody Medical AB that targets B7-H3 (CD276), an immunomodulatory glycoprotein overexpressed in various solid tumors. The drug consists of a small engineered protein scaffold, known as an Affibody molecule (approximately 7 kDa), labeled with the radioisotope lutetium-177. Upon binding to B7-H3-expressing cancer cells, the lutetium-177 emits cytotoxic beta radiation, leading to DNA damage and cell death. ABY-071 is being developed for the treatment of several high-unmet-need malignancies, including triple-negative breast cancer (TNBC), pancreatic ductal adenocarcinoma (PDAC), and both small cell and non-small cell lung cancers. Its small size is designed to facilitate rapid tumor penetration and efficient clearance from healthy tissues, potentially improving the therapeutic index compared to larger antibody-based radiopharmaceuticals.
