Overview

ABY-251 is a HER2-targeting Affibody molecule being developed as a therapeutic radiopharmaceutical for the treatment of HER2-positive solid tumors. It is a small, 7 kDa structured protein scaffold that binds to the Human Epidermal Growth Factor Receptor 2 (HER2) with high affinity (KD = 100pM). ABY-251 is designed to be labeled with rhenium-188 (188Re), a beta-emitting radionuclide, to deliver targeted radiotherapy directly to HER2-expressing tumors. It is intended for use in a theranostic approach, where patients are first screened with the diagnostic imaging agent ABY-025 to confirm HER2 expression before receiving ABY-251 therapy. Potential indications include metastatic breast cancer, non-small cell lung cancer, and gastroesophageal junction cancer.
