AC-003

AC-003 is a novel oral small-molecule inhibitor of receptor-interacting protein kinase 1 (RIPK1), developed by Accropeutics (Accro Bioscience) for the treatment of several inflammatory and fibrotic diseases. Its mechanism of action involves selective inhibition of RIPK1, a key regulator in cell death and inflammation pathways. Preclinical research suggests that AC-003 is non-immunosuppressive and may help control infection while retaining beneficial graft-versus-leukemia effects in transplantation settings. The drug has received Orphan Drug Designation from the US FDA for acute graft-versus-host disease (aGVHD). As of early 2025, it is in Phase I clinical trials for indications including acute graft-versus-host disease, idiopathic pulmonary fibrosis, rheumatoid arthritis, and ulcerative colitis.
