Overview

AC-101 is a highly potent and selective oral small molecule inhibitor of receptor-interacting protein kinase 2 (RIPK2), developed by Accro Bioscience (Suzhou) Limited for the treatment of inflammatory bowel disease, including ulcerative colitis. RIPK2 is a serine/threonine kinase involved in inflammation and innate immunity, primarily acting through the NOD/RIPK2/NF-kB or MAPK signaling pathways. By inhibiting RIPK2, AC-101 disrupts pro-inflammatory and antimicrobial responses associated with chronic inflammatory disorders. Preclinical studies demonstrated therapeutic benefits in animal models of inflammatory bowel disease, while Phase I clinical trials showed favorable pharmacokinetics, robust RIPK2 inhibition, and good safety/tolerability profiles in healthy subjects. The drug has received FDA clearance for Phase II clinical trials targeting moderate-to-severe ulcerative colitis[1][5][6][8].
