**ac-201 + febuxostat** is an investigational combination therapy consisting of AC-201, a selective oral inhibitor of TYK2 and JAK1, and febuxostat, a xanthine oxidase inhibitor. AC-201 targets cytokine signaling (notably TYK2/JAK1-dependent inflammatory pathways) and has demonstrated urate-lowering and anti-inflammatory effects, including inhibition of caspase-1 and interleukin-1β production and activity[1][2][4][6][7]. Febuxostat is primarily used as urate-lowering therapy by inhibiting xanthine oxidase, thereby reducing serum uric acid levels. The combination is being studied for synergistic urate-lowering effects and for prophylaxis against gout flares during urate-lowering therapy initiation[1][5]. AC-201 is being developed by Accropeutics (Accro Bioscience (Suzhou)), with febuxostat already widely approved.
