Overview

Janus kinase 1 is a member of the Janus kinase family of non-receptor tyrosine kinases. It plays a central role in cytokine receptor signaling by associating with type I and type II cytokine receptors. Upon ligand binding to these receptors, JAK1 becomes activated through phosphorylation events that enable it to phosphorylate STAT proteins. The activated STATs then translocate into the nucleus to regulate gene transcription involved in immune responses, cell growth, differentiation, and survival. Dysregulation or mutation of JAK1 is implicated in various diseases including cancers (such as pancreatic cancer), inflammatory conditions, infectious diseases like hepatitis D virus infection, and immune system disorders. Therapeutically targeting JAK1 has proven effective for several autoimmune diseases but carries risks related to immunosuppression.
