Ac-225-HER3-ARC is a first-in-class antibody radio-conjugate (ARC) being developed through a research collaboration between Actinium Pharmaceuticals and AVEO Oncology. The drug consists of AVEO's anti-HER3 (ErbB3) targeting monoclonal antibody conjugated with Actinium-225, a potent alpha-emitting radioisotope, using Actinium's proprietary Antibody Warhead Enabling (AWE) technology platform. HER3 is a receptor overexpressed in various solid tumors, including colorectal, gastric, breast, and non-small cell lung cancers (NSCLC). The mechanism of action involves the targeted delivery of alpha radiation to HER3-expressing cancer cells, which induces lethal double-strand DNA breaks. Preclinical studies have demonstrated potent, dose-dependent anti-tumor activity across multiple HER3-positive tumor models.
