Ac-225-PP13D3 is an investigational targeted radiotherapeutic (TRT) developed by Molecular Targeting Technologies (MTTI). It is a targeted alpha therapy (TAT) consisting of the targeting ligand PP13D3 radiolabeled with the alpha-emitting isotope actinium-225 (Ac-225). The drug is designed to treat neuroendocrine tumors (NETs) and prostate cancer by delivering high-energy alpha radiation directly to tumor cells. Alpha particles are characterized by high linear energy transfer (LET) and a very short range (typically 50–100 μm), which allows for the induction of lethal double-strand DNA breaks in cancer cells while minimizing damage to surrounding healthy tissue. The PP13D3 ligand is part of MTTI's platform designed to optimize the pharmacokinetic profile and tumor uptake of the radiopharmaceutical. As of early 2024, the program is in the preclinical stage, with activities focused on filing an Investigational New Drug (IND) application.
