AC-93253 (often studied as the salt AC-93253 iodide) is a small molecule inhibitor of the tyrosine kinase Src. It has demonstrated potent inhibitory effects on Src activity and its downstream signaling pathways, including EGFR, STAT3, FAK, PI3K, and MAPK (MEK/ERK). In preclinical models of non-small cell lung cancer (NSCLC), AC-93253 suppresses cell proliferation, migration, and invasion in vitro and inhibits tumor growth in vivo. Notably, it has been shown to sensitize both gefitinib-sensitive and gefitinib-resistant NSCLC cell lines to EGFR inhibition, suggesting potential for combination therapy strategies to overcome drug resistance in lung cancer.
