Overview

AC102 is a novel small molecule drug candidate developed for the treatment of sudden sensorineural hearing loss (SSNHL), including idiopathic sudden sensorineural hearing loss (ISSNHL). It is formulated as a specialized gel for local, intratympanic administration to the middle ear, enabling targeted delivery to the inner ear while minimizing systemic exposure. Mechanistically, AC102 acts by protecting and rejuvenating sensory hair cells and their connections to the auditory nerve. Preclinical studies have shown that it promotes recovery of hearing by reducing reactive oxygen species (ROS), enhancing mitochondrial function, preventing apoptosis in neuronal cells, and supporting neurite growth. The drug also exhibits anti-inflammatory properties by increasing anti-inflammatory factors in response to acoustic trauma. Clinical development is led by AudioCure Pharma GmbH, with support from partners such as InfectoPharm Arzneimittel und Consilium GmbH and MED-EL Medical-Electronics. As of 2025, AC102 is undergoing Phase 2 clinical trials in Europe for SSNHL[1][2][3][4][5][6][7][8].
