Overview

AC165198 is a synthetic peptide hybrid ("phybrid") developed by Biocon Ltd. as a combination therapy for metabolic diseases. It is a covalent fusion of a glucagon-like peptide-1 receptor (GLP-1) agonist (exenatide analog AC3082) linked to a second-generation amylin analog (davalintide).[1] The drug acts as a dual agonist at both GLP-1 and amylin receptors, though with slightly reduced potency at the calcitonin receptor. In preclinical studies using obese diabetic mice and diet-induced obese rats, AC165198 demonstrated greater weight loss compared to either component administered alone, with effects comparable to co-administration of both peptides. The combination approach was designed to harness complementary mechanisms for additive or synergistic metabolic benefits, including glucose control, weight reduction, and preservation of lean mass while reducing adiposity.[1]
