AC4847 is a preclinical, first‑in‑class, highly potent and selective phosphatidylinositol 3‑kinase alpha (PI3Kα)–targeting degrader being developed by Accutar Biotechnology for oncology indications driven by aberrant PI3Kα signaling. Using a chimeric/proteolysis-targeting design, AC4847 induces rapid, dose‑dependent degradation of PI3Kα protein, leading to profound suppression of downstream PI3K/AKT/mTOR pathway signaling and tumor cell growth in preclinical models.[1][8][5] Compared with the small‑molecule PI3Kα inhibitor inavolisib, AC4847 shows sub‑nanomolar potency and roughly 100‑fold greater activity, with minimal off‑target effects on other kinases, supporting its potential as a more effective and selective targeted therapy in PI3K pathway–dependent cancers.[1][8][5]
