This is a triple combination regimen of **acalabrutinib** (a second-generation Bruton tyrosine kinase inhibitor), **venetoclax** (a selective BCL-2 inhibitor), and **rituximab** (a monoclonal antibody targeting CD20). The combination is designed to provide a chemotherapy-free, targeted regimen for hematologic malignancies, most notably mantle cell lymphoma (MCL) and chronic lymphocytic leukemia (CLL). Acalabrutinib blocks B-cell receptor signaling by irreversibly binding to and inhibiting Bruton's tyrosine kinase (BTK); venetoclax induces apoptosis in B-cell malignancies by inhibiting BCL-2; and rituximab directs immune-mediated cell death by binding CD20 on B lymphocytes. The triplet has shown high response rates, durable molecular remissions, and manageable toxicity in clinical trials for treatment-naive mantle cell lymphoma[1][3].
