Drug intelligence / Profile preview
Acalabrutinib + nab-paclitaxel + gemcitabine is an experimental combination therapy consisting of three components: - **Acalabrutinib:** an orally administered, second-generation, covalent Bruton tyrosine kinase (BTK) inhibitor used in hematologic malignancies, which selectively and irreversibly inhibits BTK to disrupt B-cell receptor signaling and tumor growth[2][4]. - **Nab-paclitaxel:** an albumin-bound formulation of paclitaxel (marketed as Abraxane), which stabilizes microtubules to inhibit cancer cell division; approved for the treatment of various solid tumors, including pancreatic cancer[1][3]. - **Gemcitabine:** a nucleoside analog and antimetabolite (marketed as Gemzar), interfering with DNA synthesis and promoting apoptosis in rapidly dividing cells; used mainly for solid tumors such as pancreatic, lung, and breast cancers[3]. This triple regimen is under investigation in clinical trials as a novel approach for advanced malignancies, including pancreatic cancer, with the aim to combine targeted signaling inhibition (acalabrutinib) with established cytotoxic chemotherapy (nab-paclitaxel and gemcitabine)[1]. There is no evidence it is approved as a fixed combination; nab-paclitaxel + gemcitabine is a standard of care in pancreatic cancer, sometimes studied in triple regimens with BTK inhibitors as a research strategy.
