Acalabrutinib + vistusertib is an experimental combination therapy consisting of two small molecule inhibitors: **acalabrutinib**, a selective covalent inhibitor of Bruton tyrosine kinase (BTK), and **vistusertib**, a selective dual inhibitor of mammalian target of rapamycin complexes 1 and 2 (mTORC1/2). This drug combination was investigated primarily in relapsed or refractory B-cell malignancies, including diffuse large B-cell lymphoma (DLBCL). Acalabrutinib blocks B-cell receptor signaling through BTK inhibition, while vistusertib targets the mTOR pathway, which is critical for cell growth and survival.
