ACAM-FLU-A + adjuvant 2 is a **recombinant universal influenza A vaccine candidate** combining the highly conserved extracellular domain of the M2 ion channel protein (M2e) of influenza A with a hepatitis B core (HBc) carrier protein to enhance immunogenicity. In clinical studies, "adjuvant 2" refers to QS-21 Stimulon, an investigational **saponin-based immune adjuvant**. The combination aims to provide broad protection against all influenza A strains, including seasonal and pandemic variants like H5N1, by eliciting M2e-specific antibody responses. The vaccine was developed and initially tested by Acambis; the addition of QS-21 provided the strongest immune responses in clinical trials. The M2e-HBc platform represents a novel strategy to develop "universal" flu vaccines that do not require annual reformulation.
