Acarovac is a subcutaneous allergen immunotherapy (AIT) product developed for the treatment of house dust mite (HDM) allergy, including allergic rhinitis and asthma. The formulation contains allergoids derived from Dermatophagoides pteronyssinus and Dermatophagoides farinae (house dust mites), which are chemically modified to reduce allergenicity while retaining immunogenicity. These allergoids are adsorbed onto microcrystalline tyrosine (MCT), a biodegradable adjuvant that enhances immune response by promoting T helper type 1 cytokine production and reducing IgE formation. Some formulations also include monophosphoryl lipid A (MPL) as an additional adjuvant to further modulate the immune response. The mechanism of action involves shifting the immune profile from a Th2-dominated allergic response toward a Th1 profile with increased production of IgG4 antibodies and anti-inflammatory cytokines such as IL-10, thereby inducing tolerance to HDM allergens. Clinical studies have demonstrated significant reductions in allergy symptoms and medication use, improved quality of life scores, high patient satisfaction, and good tolerability with no serious adverse events reported[1][2][4][5][7].
