Overview

Acasunlimab is an investigational, Fc-silenced IgG1 bispecific antibody designed to elicit an antitumor immune response by conditionally activating 4‑1BB (CD137) on T cells and natural killer (NK) cells, but only when simultaneously bound to programmed cell death ligand 1 (PD‑L1). This dual mechanism aims to enhance immune-mediated tumor cell killing while minimizing off-tumor toxicity. Acasunlimab is being developed for the treatment of various solid tumors, with its most advanced indication being PD‑L1-positive metastatic non-small cell lung cancer (NSCLC), where it has shown promising results in combination with pembrolizumab in clinical trials. The drug is also under investigation for endometrial cancer and other solid tumors[2][3][7].
