Overview

ACCG-2728 is a small molecule amylin receptor agonist being developed by Structure Therapeutics for the treatment of obesity and weight management. As a non-peptide small molecule, it is designed to activate the amylin receptor, a GPCR pathway involved in appetite regulation and glucose metabolism, potentially offering an oral alternative to injectable amylin peptide analogs. Preclinical data presented at the ADA 2026 meeting evaluated the compound's gastrointestinal tolerability using a conditioned taste avoidance (CTA) model in lean rats. The results indicated that ACCG-2728 exhibits a nausea-like behavioral profile comparable to the amylin peptide cagrilintide and distinct from GLP-1 receptor agonists like semaglutide, suggesting its potential as a differentiated weight loss therapy.
