Overview

ACCG-3535 is an oral small molecule dual amylin and calcitonin receptor agonist (DACRA) being developed by Structure Therapeutics for the treatment of obesity. As a second-generation DACRA, it features a unique chemical structure distinct from the company's lead amylin compound, ACCG-2671. Preclinical data indicates that ACCG-3535 possesses high binding affinity for both human amylin and calcitonin receptors, leading to robust suppression of food intake and significant dose-dependent body weight reduction in diet-induced obese rat models. Furthermore, the compound has demonstrated enhanced weight loss efficacy when used in combination with the GLP-1 receptor agonist semaglutide. Structure Therapeutics plans to advance ACCG-3535 into Phase 1 clinical trials in the second half of 2026.
