Overview
Accum-ADC refers to a class of antibody-drug conjugates (ADCs) that utilize the proprietary **Accum** technology platform developed by **Defiante**, a subsidiary of **Scancell**. The Accum technology is designed to enhance the delivery of therapeutic payloads into the cytosol of target cells by facilitating the escape of the ADC from endosomes. Traditionally, ADCs are internalized via endocytosis and often become trapped or degraded within lysosomes; Accum-modified ADCs incorporate a specific peptide sequence that promotes endosomal membrane disruption or bypass, thereby increasing the concentration of the active drug (such as a cytotoxic agent) within the intracellular compartment. This mechanism is intended to improve the therapeutic index of ADCs by increasing potency and potentially overcoming resistance mechanisms associated with inefficient endosomal escape. Scancell is exploring the application of this platform across various oncology targets to develop more effective targeted therapies.
