AccuTOX is a novel small molecule anticancer agent developed by Defence Therapeutics. It is an optimized derivative of the parent Accum molecule and functions as both a direct tumor-killing agent and an immune system booster. Mechanistically, AccuTOX disrupts endosomal membranes in cancer cells, impairs intracellular transport mechanisms, induces genotoxic effects, blocks DNA repair pathways used by cancer cells to recover from genomic damage, and triggers immunogenic cell death. This leads to increased production of reactive oxygen species within tumor cells and enhances antigen presentation—making tumors more visible to the immune system. Notably, AccuTOX can transform "cold" tumors (with low immune infiltration) into "hot" tumors that are more susceptible to attack by immune cells. Preclinical studies have shown that it synergizes with immune checkpoint inhibitors (such as anti-PD1), resulting in improved control of solid tumor growth including melanoma, breast cancer, lung cancer, colon cancer, and T-cell lymphoma[2][3][5][6]. The drug has demonstrated efficacy across multiple murine and human cancer models with no significant toxicity observed in animal studies.
