**AccuTOX-Trastuzumab** is an experimental HER2-targeted radio-immunoconjugate being developed by Defence Therapeutics with Canadian Nuclear Laboratories through the CNRI-H program. It combines trastuzumab, which targets **human epidermal growth factor receptor 2**, with the AccuTOX endosomal-escape molecule and is intended to be radiolabeled with indium-111. Following HER2-mediated cellular uptake, the AccuTOX moiety is designed to promote escape from endosomal sequestration and nuclear localization, enabling indium-111-derived Auger electrons to induce lethal DNA damage. The program is intended for HER2-positive solid tumors resistant to established HER2-directed antibodies and antibody-drug conjugates and remains at the preclinical research stage.
