ACD137 is a small molecule drug candidate developed by AlzeCure Pharma for the treatment of severe pain conditions, particularly osteoarthritis and arthralgia. It acts as a highly selective negative allosteric modulator (NAM) of the tropomyosin receptor kinase A (TrkA), thereby blocking nerve growth factor (NGF)-mediated signaling via TrkA receptors. This mechanism has strong genetic, preclinical, and clinical validation for its role in pain modulation. Preclinical studies have demonstrated that ACD137 provides potent analgesic effects comparable to anti-NGF antibodies such as tanezumab and also exhibits significant anti-inflammatory properties. Additionally, it has shown protective effects on articular cartilage in osteoarthritis models. The compound is orally bioavailable and currently in preclinical development[1][2][6][7][8].
