ACE-054 (also known as AC0054) is a small-molecule, irreversible inhibitor of Bruton's Tyrosine Kinase (BTK) originally developed by ACEA Therapeutics and subsequently acquired by Sorrento Therapeutics in 2021. The compound is designed to target B-cell receptor signaling, which is a key driver in the pathogenesis of various B-cell malignancies, including non-Hodgkin lymphomas and chronic lymphocytic leukemia. By forming a covalent bond with the BTK enzyme, ACE-054 provides potent and sustained inhibition of downstream signaling pathways involved in cell proliferation and survival. In addition to its oncology applications, ACE-054 is being explored for its potential to treat fibrotic and autoimmune diseases, where BTK-mediated activation of immune cells like macrophages and B-cells contributes to chronic inflammation and tissue remodeling.
