Overview

ACE-2494 is a fusion protein and ligand trap developed by Acceleron Pharma for the systemic enhancement of skeletal muscle mass and strength. It functions by selectively inhibiting key negative regulators of skeletal muscle growth, including myostatin (GDF8), activin A, activin B, and GDF11—members of the TGF-beta superfamily—by sequestering these ligands in circulation. This inhibition leads to increased muscle mass and improved bone properties in preclinical models. The drug was designed to avoid effects on vascular regulators seen with other ActRIIB fusion proteins. Development was discontinued at Phase 1 due to the frequency of antidrug antibodies observed in healthy volunteers[1][2][3][5][6][7].
