**ACE527** is an investigational, orally administered, live-attenuated recombinant bacterial vaccine designed to prevent diarrhea caused by enterotoxigenic *Escherichia coli* (ETEC). It comprises three deletion-attenuated ETEC strains, ACAM2025, ACAM2022, and ACAM2027, collectively expressing the ETEC colonization antigens CFA/I, CS1, CS2, CS3, CS5, and CS6, plus the nontoxic B subunit of heat-labile enterotoxin. The vaccine is intended to elicit mucosal and systemic immune responses that inhibit ETEC intestinal colonization and neutralize toxin-associated disease. ACE527 was evaluated in Phase 1 and Phase 2 human challenge studies, including studies conducted alone and separately in co-administration with the investigational mucosal adjuvant dmLT; dmLT is not an inherent component of ACE527.
