ACE723 is an investigational, GPC3-targeted dual-payload antibody-drug conjugate (ADC) being developed by Acepodia. Engineered using Acepodia's proprietary Antibody-Dual-Drugs Conjugation (AD2C) platform, which leverages bioorthogonal click chemistry, ACE723 is designed to deliver two distinct cytotoxic payloads simultaneously to glypican-3 (GPC3)-expressing tumor cells. This dual-payload design is intended to address tumor heterogeneity and overcome potential resistance mechanisms associated with single-payload therapies. In September 2026, the US FDA cleared Acepodia's Investigational New Drug (IND) application for ACE723, enabling the initiation of a Phase 1 clinical trial to evaluate its safety, tolerability, pharmacokinetics, and preliminary anti-tumor activity in patients with unresectable or metastatic hepatocellular carcinoma (HCC).
