Drug intelligence / Profile preview
Pistabro-AB is a fixed-dose combination of acebrophylline (100 mg) and doxofylline (400 mg) developed by Pistacia Pharmaceuticals for the treatment of obstructive airway diseases, including bronchial asthma, chronic obstructive pulmonary disease (COPD), and acute or chronic bronchitis. Acebrophylline is a xanthine derivative that combines theophylline-7-acetic acid with ambroxol, acting as both a bronchodilator and a mucolytic agent. It reduces mucus viscosity, enhances mucociliary clearance, and exerts anti-inflammatory effects by inhibiting phospholipase A2 and reducing the production of pro-inflammatory mediators like leukotrienes. Doxofylline is a novel methylxanthine bronchodilator that inhibits phosphodiesterase (PDE) enzymes, specifically PDE2A and PDE4, leading to increased intracellular cyclic AMP (cAMP) levels and subsequent relaxation of bronchial smooth muscle. Unlike traditional xanthines like theophylline, doxofylline has minimal affinity for adenosine receptors, resulting in a significantly improved safety profile with fewer cardiac and gastrointestinal side effects.
