Overview

Acemannan is a highly acetylated mannan polysaccharide derived from Aloe vera, specifically Aloe barbadensis Miller. It is the primary bioactive component in Aloe vera gel and consists of D-isomer mucopolysaccharides linked by β-(1,4) glycosidic bonds. Acemannan exhibits immunomodulatory, antiviral, antitumor, osteogenic, anti-inflammatory, and antibacterial properties. Mechanistically, it induces cytokine production (including interferon gamma, tumor necrosis factor-alpha, and interleukins), activates macrophages via Toll-like receptor 5 (TLR5)/NF-kB signaling pathways in gingival fibroblasts, and promotes tissue repair through AKT/mTOR-mediated protein translation. Clinically investigated for oral diseases such as aphthous ulcers and candidiasis (e.g., Celagace Orarinse Solution), it also shows potential for wound healing and bone regeneration[1][2][3][5][6]. In veterinary medicine it has been used to manage fibrosarcoma in dogs and cats[5].
